NeuronSifter: An Agent-in-Twin for Prioritizing Central Nervous System Interventions with World Action Models

NeuronSifter is a decision‑centric Agent‑in‑Twin framework that uses an evolving biological digital twin instead of a fixed predictive model to prioritize central nervous system interventions. It compiles interventions into state‑conditional target‑occupancy representations, propagates them through a mechanism‑grounded World Action Model, and selects informative measurements to reduce decision uncertainty. The system is evaluated on Alzheimer’s disease scenarios, demonstrating adaptive evidence acquisition, uncertainty‑aware planning, and robustness to model discrepancy, while continuously refining its decision‑relevant representations with externally verified evidence.

Transition-Based Digital Twin Modelling for Alzheimer's Disease under Sparse Longitudinal Data

Alzheimer's disease (AD) progression is highly heterogeneous and is typically observed through sparse and irregular longitudinal data, posing challenges for prediction and personalised monitoring. Existing machine learning approaches have improved AD prediction using multimodal data, yet often focus on static classification or cohort-level risk estimation, providing limited support for subject-specific modelling and uncertainty-aware reasoning.

NeuronSifter: Intervention Planning in CNS Microenvironments

NeuronSifter is a framework for planning interventions in central nervous system microenvironments by converting treatment regimens into state‑conditional target‑occupancy fields and propagating them through microenvironment dynamics. It selects measurements based on their expected reduction in intervention loss, integrating typed outcomes into a unified posterior. In synthetic Alzheimer’s disease scenarios, occupancy conditioning improves trajectory probability scores and intervention ordering accuracy, and decision‑directed acquisition reduces terminal risk compared to a Bayesian experimental design planner.

Graphical conditional generative modeling for digital twin modeling

arXiv:2606. 16219v1 Announce Type: cross Abstract: Digital twin modeling, including control and data assimilation under model uncertainty, often faces an open-ended fidelity problem: adding variables, data streams, and time scales can indefinitely increase model complexity, ultimately producing systems that are difficult to maintain, validate, interpret, and use for stress or safety testing.
