Imaging-anchored multiomics in cardiovascular disease: integrating cardiac imaging, bulk, single-cell, and spatial transcriptomics

Read the original on arXiv AI →

The article reviews how cardiac imaging techniques such as echocardiography, CMR, and CT can be integrated with bulk, single-cell, and spatial transcriptomics to create joint representations that link imaging phenotypes to detailed molecular states. It outlines the representation needs of each modality, compares multimodal fusion strategies, and synthesizes pipelines for radiogenomics, spatial alignment, and image-based gene‑expression prediction, while discussing validation, limitations, and failure modes. The review highlights emerging spatial multiomic maps of human myocardium and atherosclerotic plaque, noting that cost, scalability, and tissue availability remain major barriers to large‑scale cardiovascular translation.

Biology-Guided Context-Aware Residual Propagation for Spatial Gene Expression Prediction from Histology

The paper introduces BCARP, a Biology‑Guided Context‑Aware Residual Propagation framework that predicts spatial gene expression from H&E‑stained histology images. Rather than directly decoding expression from visual features, BCARP treats histology embeddings as morphological priors and iteratively refines spot representations in a transcriptomics‑oriented latent space, integrating nuclei‑guided local context and prototype‑guided global propagation. Evaluations on three spatial transcriptomics datasets and ten HEST‑1k cohorts show that BCARP outperforms existing baselines in mean correlation‑based metrics, demonstrating the effectiveness of biology‑guided local‑global residual propagation for histology‑to‑transcriptomics prediction.
